Gibberellic Acid-induced Phase Change in Hedera helix as Studied by Deoxyribonucleic Acid-Ribonucleic Acid Hybridization.
Applications of gibberellic acid to the mature form of Hedera helix induce morphological reversions to the juvenile form of growth. The juvenile forms produced are stable with time and differ dramatically from the mature in phenotype. DNA-RNA hybridization techniques have been used to study the RNA populations of juvenile, mature and gibberellic acid-treated mature apices. Hybridization competi...
متن کاملAuxin and ribonucleic Acid synthesis in pea stem tissue as studied by deoxyribonucleic Acid-ribonucleic Acid hybridization.
The ability of auxin to alter gene transcription in pea (Pisum sativum L.) stem tissues has been investigated by means of DNA/RNA hybridization-competition techniques. In order to obtain reproducible hybridization with total nucleic acid preparations from plants it was found necessary to remove interfering substances, probably polysaccharides; this was accomplished by methoxyethanol extraction ...
متن کاملPrinted in Great Britain Ribonucleic Acid-Deoxyribonucleic Acid Hybridization as a Second-Order Reaction By
The formation of specific RNA-DNA hybrid molecules has been widely used to study relationships between RNA and DNA sequences. Although this reaction probably involves mechanisms similar to those of DNA renaturation, the analysis of its kinetics is more complicated because the relative concentrations of complementary RNA and DNA sequences are often unknown. Moreover during hybridization the DNA ...
متن کاملRibonucleic acid-deoxyribonucleic acid hybridization in aqueous solutions and in solutions containing formamide.
Hybridization in 6xSSC (SSC, 0.15m-sodium chloride-0.015m-sodium citrate) at 66 degrees C was compared with hybridization in formamide-6xSSC (1:1, v/v) at 35 degrees C. As expected, the RNA hybridization potential was labile in the former system and stable in the latter. DNA retention by filters was poor in the formamide system, but could be improved. Several other properties of the hybridizati...
متن کاملDeoxyribonucleic acid-deoxyribonucleic acid hybridization of R factors.
A new method of estimating deoxyribonucleic acid homology is presented and used to measure the homology between bacterial plasmids of different compatibility groups. Deoxyribonucleic acid of the P-group plasmid RP1, originally isolated in Pseudomonas aeruginosa, has considerable (50%) homology with an N-group plasmid of the Enterobacteriaceae.
